Automated Library Preparation for NGS

DBS-LibPrep offers simplified operations, significantly reduced reagent usage, and the ability to produce high quality libraries.

As long as 1/5, 1/10 or even 1/20 of the samples and reagents used in traditional library construction, DBS-LibPrep can automatically complete fragmentation, end repair and A-tailing, adapter ligation, library amplification (PCR), and bead cleanup. Users only need to add samples and reagents, and other steps are automatically completed on the chip. The KAPA library preparation can be completed in 1.5 hours. DBS-LibPrep is flexible and suitable for different library preparation methods.

The following is the flowchart of running KAPA HyperPlus Library Prep Kit on DBS-LibPrep. Currently, the reaction volumes have been reduced to 5%, but the reaction steps and times are kept the same as the original protocol. Optimization will be done next to reduce the overall library prep time.





Instrument Operations:

A touch screen is provided to make the instrument easy to use. Small volumes of samples and reagents are loaded to a DMF chip. All library preparation steps are performed on the DMF chip in a DBS-LibPrep instrument. Only 5 minutes of hands-on time is needed to set up an experiment.

Reaction volume: 1/20 of the recommended volume in the Kit

Reaction time:Same as the recommended in the Kit

Experiments are carried out to verify the performance of using a digital microfluidic chip for library preparation, in which the reaction volume is reduced to 1/20 of the conventional volume. Reaction time is kept the same for easier comparison purposes. The input DNA (12.5ng) is provided by a collaborator. On-chip and off-chip performance results are compared for all the individual library preparation steps. The Agilent 2100 measurements show good consistency between on-chip and off-chip library preparation.
